Description	The aim of the work was to optimize the most important steps in a preparation of molecular cytogenetic slides for SKY analysis. The optimization included: looking for optimal protease concentration, pretreatment and its comparison, aging types influence on chromosome structure and shape. The result was a suggestion of optimal protocol for SKY. Practical possibilities of SKY were shown on material from cytogenetic investigation.
